These antibiotics prevent the growth of the bacteria by inhibiting certain steps in the biosynthesis of purines and therefore nucleic acids which is necessary for DNA replication.
Both Sulfamethoxazole and Trimethoprim act as a competitive inhibitor of dihydropteroate synthetase, a bacterial enzyme. These two compounds hinder the combination of Aminobenzoic acid (PABA) and dihydropteroate synthetase. For the synthesis of folic acid to take place, this inhibition reaction is necessary. As a result of this, the bacteria are unable to produce folic acid, which is essential for its multiplication.
